چکیده

Autophagy is a major catabolic process in eukaryotes that help degrade and recycle macromolecules and organelles. Recent evidences suggest autophagy to mediate cytoprotection and also help increase longevity. It acts as a central regulatory mechanism for aging/longevity in diverse eukaryotic species. In the present study we have exploited the lower eukaryotic model organism, Dictyostelium discoideum to study longevity, which could use autophagy as a major mechanism for increasing lifespan. This organism is unique as it follows a developmental programmed cell death by autophagy that is independent of caspases. It could be a beneficial organism to delineate the mechanism of increased longevity that follows autophagy.

Analysis of rapamycin induced autophagy in Dictyostelium discoideum.

Natural autophagy and autophagic cell death is being studied in the model system, D. discoideum, which has well known genetic and experimental advantages over the other known systems. There is no apoptotic machinery present in this organism which could interfere with the non-apoptotic cell death. The target of rapamycin (TOR) pathway is a major nutrient-sensing pathway which when inhibited by t...

A proteolytic cleavage assay to monitor autophagy in Dictyostelium discoideum.

Dictyostelium discoideum is a good model of autophagy. However, the lack of autophagic flux techniques hinders the assessment of new mutants or drugs. One of these techniques, which has been used successfully in yeast and mammalian cells, but has not yet been described in Dictyostelium, is based on the presence of proteolytic fragments derived from autophagic degradation of expressed fusion pro...

Methods to Monitor and Quantify Autophagy in the Social Amoeba Dictyostelium discoideum

Autophagy is a eukaryotic catabolic pathway that degrades and recycles cellular components to maintain homeostasis. It can target protein aggregates, superfluous biomolecular complexes, dysfunctional and damaged organelles, as well as pathogenic intracellular microbes. Autophagy is a dynamic process in which the different stages from initiation to final degradation of cargo are finely regulated...

Eat Prey, Live: Dictyostelium discoideum As a Model for Cell-Autonomous Defenses

The soil-dwelling social amoeba Dictyostelium discoideum feeds on bacteria. Each meal is a potential infection because some bacteria have evolved mechanisms to resist predation. To survive such a hostile environment, D. discoideum has in turn evolved efficient antimicrobial responses that are intertwined with phagocytosis and autophagy, its nutrient acquisition pathways. The core machinery and ...
